sp7 the secret gene for bone growth
SP7 is a crucial protien gene locatedn in human chromosomes 12 (12q13) that acts as a master regulator of osteoblast differentation and bone formation.
SP7's function:
Sp7 acts as a master regulator of bone formation during both embryonic development and during the homeostatic maintenance of bone in adulthood.

A GWAS study has found that bone mass density (BMD) is associated with the Sp7 locus, adults and children with either low or high BMD were analyzed showing that several common variant SNPs within the 12q13 region were in an area of linkage disequilibrium

There are two main pathways which cause induction of sp7 gene, Msx whic hdirently contributs to sp7 where as bone morphogenetic protein BMP2 contributs to it indirectly through Runx2 study link

during development

In a developing human embryo, Sp7 acts as one of the most important regulatory shepherds guiding bone formation. The process of creating solid ossified bone begins with mesenchymal stem cells differentiating into chondrocytes, after which some of those chondrocytes turn into cartilage. Certain areas of this early cartilage then serve as a natural template for bone cells while the whole skeleton continues to develop.
In adults:
in adult mice ablation of sp7 led to a lack of new bone formation (less bone formation) highly irregular cartilage accumaliation beneath growht plate and defects in osteocyte maturation and funciontality study link
Other studies have shown that when Sp7 is conditionally knocked out in the osteoblasts of adult mice, the animals develop osteopenia in their vertebrae.
This highlights how Sp7 continues to play a vital role in maintaining healthy bone mass even after skeletal development is complete.
Clinical relevance:
Osteoporosis GWAS studies show assocation between adult and juvenile bonemass desnity (BMD) and sp7 in humans, though low BMD a good indicator for osteoporisis in adults there are no corelation between the two.
Rheumatoid Arthritis:
is a protein hormone that has beens shown to upregulate in rheuumatoid arthsits pathology, , causing the release of inflammatory cytokines and enhancing the breakdown of the bone matrix.
